Summary:A biological material imaging calibration device and method of forming such. A first set of discrete regions are provided upon a retaining member, each region containing a selected tissue stain material, which exhibits a predetermined optical response inside a colour-stable region of a chromaticity diagram, and providing a second set of discrete regions formed of coloured filters having predetermined optical responses outside the colour stable region. The selected tissue stain material may produce stable results over a year or 100 calibrations. Region of the first set may comprise an absorbent material stained with a respective material. The first set of regions may be provided by staining biopolymer films. The first set of regions may be provided by machining or casting recess areas. The first set of regions may be provided by a masking element with cut out regions. The first set of regions may be provided by printing the tissue stain materials onto a surface. Stain materials in different regions may have different concentrations. Providing the first set of regions may comprise spin coating the retainer with tissue stain materials. Providing the first set of regions may comprise depositing a thin film onto the retainer.
