Summary: | Provided are compositions and methods that include an engineered anti-CRISPR (Acr) protein that can achieve a balance in which Cas proteins retain activity to perform on-target nucleic acid targeting functions (e.g., DNA cleavage for gene editing applications), but are inhibited by a subject Acr protein to a degree that decreases off-target activity - thus resulting in an increased ratio of on-target to off-target nucleic acid targeting events. In some cases, a subject composition (e.g., a subject system) includes a CRISPR nuclease or a nucleic acid encoding it. Methods of using a subject Acr polypeptide together with a CRISPR nuclease to achieve target nucleic acid modification are provided. |
