The present invention relates to a pharmaceutical composition for preventing and treating cancer, containing cytochrome b5 reductase 3 (cyb5R3) protein as an active ingredient. The expression of hypoxia-inducible factor-1 (HIF-1) is significantly reduced in cancer cells over-expressing cyb5R3 so as to inhibit the growth of cancer cells and effectively inhibit in vivo tumor growth and metastasis, and thus the cyb5R3 gene or protein of the present invention can be useful as an active ingredient of a pharmaceutical composition for preventing and treating cancer.
