Summary:The following is claimed: a process for the alignment of macromolecule(s) on the surface (S) of a support, characterised in that the triple line S/A/B (meniscus) resulting from the contact of a solvent (A) with S and a medium (B) is displaced on S, the macromolecules having one part, esp. one end, anchored to S, the other part, esp. the other end, being in solution in A. Also claimed are:(a) a surface with aligned macromolecule(s) obtained by the above process; (b) a process for the detection, sepn. and/or determn. of a macromolecule in a sample, comprising the use of an alignment process as above in which a biologically active mol. capable of recognising the target macromolecule is fixed to S, and the use of an opt. fluorescent reagent to detect the fixed mol. or macromolecule;(c) a process for physically mapping a gene in genomic DNA, where the DNA is aligned and/or detected as above;(d) a kit for carrying out process (c), comprising the total genomic DNA of a reference host, a support with a surface capable of anchoring and aligning the DNA, probes specific for the gene to be mapped, and reagents for the hybridisation and detection of the DNA;(e) a process for diagnosing a pathology associated with the presence or absence of a DNA sequence specific to the pathology (see `Claimed Process');(f) a kit for carrying out process (e), comprising a support with a surface capable of anchoring and aligning the patient's DNA, probes specific to the gene implicated in the pathology, and reagents for the hybridisation and detection of the DNA;(g) a kit for carrying out process (e), comprising probes as in (f) anchored and aligned on the surface of a support, reagents for labelling the DNA, esp. the patient's DNA, and reagents for the hybridisation and detection of the DNA;(h) a process for the prepn. of a gene from genomic DNA, comprising identifying the position. of the gene on the DNA by a process as above using a probe specific to the gene, enzymatically amplifying the gene sequence and/or its flanking sequences, and isolating the amplified prod.;(i) a DNA construct contg. a gene prepd. by process (h), opt. associated with a homologous or heterologous regulatory sequence;(j) use of a gene obtained by process (h) to prepare a DNA construct useful in a process for replacing a gene in the genome of a eukaryotic cell by targeted insertion of a foreign gene, where the foreign gene is prepd. by process (h), and(k) a vector useful in a gene replacement process as in (j), where the foreign gene is prepd. by process (h).
