Summary: | The present invention provides a method for increasing expression of low molecular weight polypeptides which are produced by recombinant DNA tranformation of prokaryotic host cells. This invention allows the direct expression of these polypeptides without fusion to an otherwise expressable protein and requires only the addition of a single amino acid to the sequence of the polypeptide product of interest. The method of producing a polypeptide derivative which comprises the structure Methionine-X-R wherein X is an amino acid selected from the group consisting of Alanine, Arginine, Glutamine, Glycine, Isoleucine, Leucine, Lysine, Methionine, Phenylalanine, Serine, Threonine, Tryptophan, and Valine, and R is the amino acid sequence of any polypeptide product of interest; in a prokaryotic host cell which has been transformed with a recombinant DNA vector, said vector comprising; A. a DNA sequence that provides for autonomous replication or chromosomal integration of said vector in said host cell; B. promoter and translational activating sequence functional in said host cell; and C. a DNA compound which comprises the coding sequence of a polypeptide derivative described herein, positioned in transcriptional phase with said promoter and translational activating sequence; said mothod comprising culturing said prokaryotic host cell under conditions suitable for gene expression. |
