Summary:The invention relates to purified thermostable DNA polymerases from Pyrodictium species, such as Pyrodictium occultum or Pyrodictium abyssi, which polymerases catalyze the combination of nucleoside triphosphates to form a nucleic acid strand complementary to a nucleic acid template strand. The preferred polymerases are characterized by their ability to function efficiently in a polymerase chain reaction, wherein said reaction includes repeated exposure to a denaturation temperature of about 100 DEG C. Most preferably the polymerases display 5'->3' exonuclease activity, i.e. are proofreading enzymes. The invention also provides DNAs encoding the DNA polymerase activity of the said Pyrodictium species, which DNAs can be used to construct recombinant vectors and transformed host cells for production of polypeptides having said activity. The invention also relates to the preparation of said thermostable DNA polymerases, to the use of said polymerases to amplify nucleic acids as well as to kits comprising a polymerase of the present invention.
