Summary:The present invention relates to a method of deforming the identity of one or more nucleotide bases of a plurality of specific positions in one or more nucleic acid molecules of interest. The method involves the provision of a single-stranded nucleic acid, which is contacted with hybridization printers to form duplexes that are contacted with a terminator reagent and undergo extension and affinity separation. This enables one or more nucleotide bases of a plurality of specific positions in one or more nucleic acid molecules of interest to be determined.
