Culturing Rhodococcus rhodochrous for preparation of nitrile hydratase, useful for converting nitriles to amides, particularly preparation of acrylamide, by growing in phosphate-buffered medium of pH 5.5 to 9

Method for culturing the nitrile hydratase (I)-producing strain Rhodococcus rhodochrousM33 (DSM 14230) using a medium based on 12-60 mM phosphate buffer that (a) satisfies the cells' requirement for phosphorous and (b) maintains pH 5.5-9 during culture. An independent claim is also included for...

Summary:Method for culturing the nitrile hydratase (I)-producing strain Rhodococcus rhodochrousM33 (DSM 14230) using a medium based on 12-60 mM phosphate buffer that (a) satisfies the cells' requirement for phosphorous and (b) maintains pH 5.5-9 during culture. An independent claim is also included for a medium for the process comprising, per l, 7.9 g sodium dihydrogenphosphate dodecahydrate; 1.8 g potassium dihydrogenphosphate; 0.02-0.04 g cobalt(II) chloride hexahydrate; 1 g magnesium sulfate heptahydrate; 0-10 g maize extract; 20-90 g glucose and 4-20 g urea.
