Its advantages are high safety, easy operation and low cost. 一种鱼类胚胎玻璃化冷冻保存方法,通过比较不同抗冻剂的毒性效应和玻璃化性能,筛选出适宜海水鱼类胚胎的玻璃化液配方;确定适宜玻璃化处理的胚胎发育时期,研制出特定的洗脱溶液,采用分步平衡、快速降温和解冻的技术路线,建立了鱼类胚胎玻璃化冷冻保存技术。对牙鲆不同发育时期胚胎进行了8次玻璃化冷冻实验中共获得冷冻复活胚20粒,最高复活率达32%。其中5次实验中获得冷冻复活胚胎发育孵化出鱼苗的结果。在20枚冷冻复活胚胎中,有14枚冻胚孵化出鱼苗,孵化率高达70%。冻胚孵出的鱼苗,胚体透明,形态正常,无畸形。本发明技术无需昂贵的仪器设备,操作简单,成本低廉,易于推广,可用于牙鲆等海水鱼类胚胎超低温(-196℃)冷冻保存。
