Summary: | The present invention relates to fermentation process of producing aureomycin and is the technology of shortening aureomycin fermenting period. The culture medium is prepared with corn starch, corn slurry, (NH4)2SO4, KH2PO4, MgSO4 and amylase as well as water soybean oil and defoaming agent and through mixing, sterilizing at 121 deg.c for 25 min and other steps. The culture medium has lower viscosity, increased dissolved oxygen, strengthened aureomycin growth and antiseptic synthesizing metabolism, raised spore germination rate and shortened fermentation period, and these result in low power consumption, high apparatus utilization and low cost.
本发明涉及发酵法生产四环素族金霉素技术,特别是针对缩短金霉素发酵周期的技术。(1)培养基:玉米淀粉6-7.5、酵母粉0.1-0.5、蛋白胨1.0-1.8、黄豆饼粉3-5、玉米浆1-1.5、(NH#-[4])#-[2]SO#-[4] 0.3-0.8、KH#-[2]PO#-[4] 0.015-0.02、MgSO#-[4]0.02-0.03淀粉酶0.08-0.2(20000u/g);(2)以水∶豆油∶泡敌=30-70∶10-40∶7-17其用量为0.1-0.3%。先将以上基础料中的固型物如黄豆饼粉和玉米淀粉、酵母粉用少量水搅拌均匀,无结块,再加入无机盐,并用水稀释5.5倍,加入淀粉酶和消泡剂,定容,灭菌,121℃,保持25分钟。本发明采取以上技术后,培养基粘度降低,增加了溶氧,加强了金霉素产生菌的生长和抗生素合成代谢,孢子萌发率由58%提高到82%,发酵周期由140-150小时缩短到90-100小时。其最终效益表现在大幅度降低能耗,提高设备利用率,减少管理费,成本下降42%。 |
