The invention provides a preparation method of C5 phosphonate, which comprises the following steps: carrying out oxidation reaction on C5 aldehyde to obtain C5 acid; carrying out esterification reaction on the C5 acid to obtain C5 ester; and carrying out phosphine esterification reaction on the C5 ester in the presence of a metal halide and an alkaline compound to obtain the C5 phosphonate. According to the method, the raw material C5 aldehyde for synthesizing vitamin A is used as a starting raw material, the target product can be obtained only through three-step reaction, the raw materials are easy to obtain, the reaction route is short, the reaction condition is mild, meanwhile, high reaction selectivity and high raw material conversion rate are achieved, and therefore high-yield and high-purity C5 phosphonate is obtained.
