The construction method comprises the following steps: heterologously expressing sulfur quinone oxidoreductase Sqr in sulfur alkali vibrio cells, and knocking out polysulfide transmembrane transporter protein YilD and sulfur oxidase SoxB to obtain the engineering bacterium for strengthening the sulfur oxidation function. Wherein the expression of the sulfur quinone oxidoreductase Sqr can improve the rate of oxidizing sulfide into elemental sulfur, and the knockout of the polysulfide transmembrane transporter protein YilD and the sulfur oxidase SoxB contributes to weakening the continuous oxidation of the elemental sulfur into sulfate radicals, so that the yield of the elemental sulfur is improved.
