Summary:Relates to a CRISPR-Cas13 (clustered regularly interspaced short palindromic repeats-associated protein 13) system and application thereof, and also relates to Cas13 protein, fusion protein and guide polynucleotide. The Cas13 protein has at least 90% sequence identity compared with SEQ ID NO: 1. The fusion protein comprises the Cas13 protein fused to a protein domain and/or a polypeptide tag. In some embodiments, the guide polynucleotide comprises an isotropic repeat sequence having at least 70% sequence identity to any of SEQ ID NO: 3, 80-87 and a guide sequence engineered to hybridize to the target RNA.
