The method is characterized in that mixed sulfhydrylated heparin and protamine form a compound, chloroauric acid is reduced in the compound through a hydrothermal method, and gold nanoclusters grow in situ. The heparin-protamine compound has triple functions of forming a stable dispersion system to enable the gold nanocluster growing in the heparin-protamine compound to have good water solubility, reducing chloroauric acid and forming a gold-sulfur bond anchored gold nanocluster, and selectively degrading the heparin-protamine compound by trypsin to cause fluorescence change of the gold nanocluster.
