The invention provides a hepatotoxicity risk assessment method for non-steroidal anti-inflammatory drugs and metabolites thereof based on network architecture analysis. The method comprises the following steps: collecting common non-steroidal anti-inflammatory drugs and metabolites thereof; collecting a drug target and a hepatotoxicity risk gene; establishing a non-steroidal anti-inflammatory drug target network and a hepatotoxic disease module; according to the method, hepatotoxicity risks of non-steroidal anti-inflammatory drugs and metabolites thereof are sequenced by three network-based hepatotoxicity risk assessment strategies, including network proximity, network diffusion and aggregation sequencing methods; an overlapping network of a drug target network and a hepatotoxic disease module is constructed, and a hepatotoxic molecule induction mechanism of the non-steroidal anti-inflammatory drug is explored.
