Summary:The invention discloses a method for improving the fermentation yield of ergothioneine through an overexpression transport protein coding gene enhanced product efflux process. Transport protein coding genes MRH1, PHM7, HNM1, TPO5 and SCW10 which possibly participate in ergothioneine efflux in saccharomyces cerevisiae are obtained through analysis and screening based on transcriptomics. The saccharomyces cerevisiae engineering strain is constructed by constructing a recombinant vector for over-expressing the key gene and introducing the recombinant vector into an ergothioneine-producing saccharomyces cerevisiae starting strain. Research results show that after the key gene is over-expressed, the growth of yeast engineering strain thalli is obviously improved, the fermentation yield of ergothioneine is obviously increased, and the extracellular ergothioneine ratio is obviously increased.
