The invention provides a PM/Scl antibody joint detection kit and method, the preparation method comprises: preparation of a magnetic particle reagent and preparation of an enzyme marker reagent, and the magnetic particle reagent comprises magnetic particles coated with a PM/Scl-75 antigen and a PM/Scl-100 antigen. According to the present invention, the detection kit can significantly improve the overlap syndrome sample which is missed due to the use of the single antigen as the detection raw material, and can detect the majority of the false negative samples and the samples with low signal values generated due to the missed detection, and the negative samples are not interfered. A detection platform involved in the invention is a chemiluminescence platform, and compared with a traditional immunoblotting method, the chemiluminescence detection method has the characteristics of high automation degree, wide linear range, high sensitivity, good result repeatability, more accurate quantitative result and the like
