Summary:The present invention relates to methods for producing nucleic acid-encoded compounds and libraries of nucleic acid-encoded compounds. A neogenic compound comprising a scaffold attached to a solid support by a linker is covalently attached to one or more chemical structural units to form a chemical moiety attached to the scaffold. A coding oligonucleotide encoding the one or more chemical structural units is covalently linked to the neogenic compound to form a coding nucleic acid moiety linked to the scaffold. A cleavage group is linked to the chemical moiety, nucleic acid moiety or scaffold of the compound. The linker is then reacted with the cleavage group such that the linker is cleaved and the compound is released from the solid support.
