The invention provides a preparation method of polysubstituted olefin. According to the preparation method, a strategy of firstly forming an alkenyl metal species from alkyne halide and finally performing carbon-hydrogen bond activation under the promotion of amide to form the polysubstituted olefin is developed for the first time, so that alkenyl fragments are introduced into amide molecules in a site-selective and stereoselective manner to form the polysubstituted olefin. The method comprises the following steps: under the condition of an inert solvent, under the combined action of a transition metal catalyst and an oxidizing agent, alkyne halide firstly forms allene ketone, then an alkenyl metal species is generated under the action of metal, and then a carbon-hydrogen bond activation strategy is performed to form the polysubstituted olefin. Amide and olefin are taken as ubiquitous functional groups in natural products, medicines and materials, and the reaction is expected to provide a new thought for deve
