Summary:Herein is reported a method of humanization of non-human antibodies using a structure-based scoring matrix. Using this scoring matrix, the (claim and) suitability of specific (reply) mutations of amino acid residues at defined positions of the selected human germline sequence can be determined. The scoring matrix takes into account the topology, three-dimensional structure, and interaction and variation of the corresponding residues. Thus, the effect of a specific amino acid residue change on antigen binding can be determined. 本文报道,使用基于结构的评分矩阵进行非人抗体的人源化的方法。利用该评分矩阵,可以确定在所选人种系序列的限定位置上氨基酸残基的特定(回复)突变的(要求和)适宜性。该评分矩阵考虑相应残基的拓扑学、三维结构以及相互作用和变化。因此,可以确定特定氨基酸残基改变对抗原结合的影响。
