According to the regional biomineralization method, a laser engraving technology is used for cutting a substrate to form a mask with a special hollow structure, after the mask is tightly attached to a substrate layer, negative electricity modification is conducted on the substrate layer, and the substrate layer is soaked in improved simulated body fluid, so that hydroxyapatite similar to a bone matrix can be formed in a limited region of the substrate layer; the micro-fluidic bone chip is constructed by adopting the regional biomineralization method, and the chip comprises a control layer, a flowing layer and a limited region biomineralization substrate layer which are sequentially sealed from top to bottom, so that the construction of the micro-fluidic b
