Summary: | The invention discloses a preparation method of an MOFs encapsulated enzyme for separation and analysis of low-molecular-weight protein, and belongs to the technical field of immobilized enzyme preparation. The surface of trypsin (Try) is modified with polyacrylic acid (PAA) to synthesize a Try-PAA compound, the Try-PAA compound and a ZIF-L precursor (zinc nitrate hexahydrate and 2-methylimidazole) aqueous solution are incubated together, Zn < 2 + > is attracted through PAA which is crosslinked with Try and has negative electricity, and thus Try is packaged in ZIF-L. According to the method, the defect of low efficiency is overcome, the Try is packaged in the ZIF-L in situ through a biomimetic mineralization method, and Try leakage is effectively prevented. The prepared hydrophobic mesoporous Try-PAA-ZIF-L is applied to enzymolysis analysis of LWPs in a model protein mixture and a human serum sample, and shows excellent size selective enzyme digestion performance.
