Summary: | The present invention provides a method for preparing a polysaccharide-protein conjugate, in which the polysaccharide-protein conjugate is prepared under conditions that improve conjugation reaction consistency, increase protein consumption during the conjugation reaction, produce a higher molecular weight conjugate, and/or reduce the content of free cyanide in the conjugation reaction product. Polysaccharides, typically from bacteria, are conjugated to a carrier protein by reductive amination. The polysaccharide-protein conjugates obtained using these methods can be used for inclusion in multivalent vaccines.
