Summary:The invention belongs to the technical field of gene editing, and relates to a deaminase mutant and a base editor constructed based on the deaminase mutant. A novel cytidine deaminase mutant is developed, the cytidine deaminase mutant is fused to different sites of mutant nuclease represented by SpCas9, and the obtained novel fusion protein can achieve effective C-T base mutation on cytosine located at different positions of a front spacer sequence. The Cas-independent DNA off-target effect of the fusion protein obtained by the method is close to the background level, and the fusion protein has diversified editing activity windows.
