Summary:The invention relates to a piezoelectric crystal-based biochip and a preparation method thereof. The preparation method comprises the following steps: S1, forming a detection sample reaction area from a sensitive functional area of a piezoelectric crystal sensor chip through an isolation device; s2, covering a sensitive functional area of the piezoelectric crystal sensor chip with a layer of polymeric membrane; and S3, modifying a binding carrier capable of forming specific binding with a detection object on the polymeric membrane, thereby completing the preparation of the biochip. The binding vector comprises a functional group, a nucleic acid aptamer, an antibody or a protein molecule. The problems that in the prior art, a piezoelectric crystal biosensor is complex in preparation, high in requirement, difficult in quality control, difficult in batch production and the like are solved, and the biochip which is simple in process, controllable in quality, good in effect and capable of being produced in batches
