The invention discloses a method for remarkably promoting soluble expression of expansive protein TgS and application of the method. The soluble expression of the expansive protein TgS is improved by utilizing the hydrotropy tag protein and changing and optimizing the fermentation conditions in the protein expression process. Based on an earlier-stage research result and current soluble protein utilization optimization requirements, it is found that in the protein expression process of an escherichia coli system, soluble expression of the expansive protein TgS can be achieved under the conditions that TRX tag protein is at the low temperature (12 DEG C), the ventilation capacity is 0.5 (V/V.min <-1 >) and 0.02 g.L <-1 > ZnSO4 and 0.1% glycerol are added. The protein fermentation process is combined and optimized from the inclusion body generation characteristics, soluble efficient expression during heterologous expression of the expansive protein TgS is achieved, the biological activity of the expansive prote
