The invention discloses an operating device for a valve material cell migration experiment and a using method thereof.The operating device comprises a culture plate and a pressurizing plate, a culture groove is formed in the culture plate, a plurality of protruding columns are arranged on the bottom face of the pressurizing plate, contact pads are arranged at the ends of the protruding columns, and a pressurizing device is arranged on the top face of the pressurizing plate; an anti-sticking plate is arranged between the pressurizing plate and the culture plate, a plurality of penetrating holes allowing the protruding columns to penetrate through are formed in the anti-sticking plate, the cross section size of the penetrating holes is equal to that of the protruding columns, a spring is connected between the anti-sticking plate and the pressurizing plate, two locking hooks are symmetrically arranged on the two sides of the pressurizing plate, and the locking hooks are rotationally connected with the pressurizi
