The preparation method comprises the following steps: mixing hydrophobic quantum dots, an organic non-coordination solvent and a first emulsifier solution to obtain a quantum dot micelle dispersion liquid; mixing a monomer, a ligand and a second emulsifier solution to obtain a monomer droplet dispersion liquid; and polymerizing the quantum dot micelle dispersion liquid and the monomer droplet dispersion liquid in the presence of an initiator. A ligand is added into a monomer and then polymerized with quantum dots, dynamic balance of the ligand on the surfaces of the quantum dots is achieved through the introduced ligand, high PLQY of the quantum dots is maintained, and preparation of the high-PLQY quantum dot nanospheres is achieved.
