The invention further discloses a preparation method of the Corynebacterium glutamicum. The yield of the L-serine produced by the corynebacterium glutamicum reaches 33.7 g/L and is increased by 10.86% compared with that of an original strain, and after the alpha-ketoglutaric acid is added, the biomass is not greatly changed, the yield of the L-serine is increased by 36.2%, and the efficiency of producing the L-serine by using the corynebacterium glutamicum is greatly improved.
