Summary:Compositions and methods for modifying genomic DNA sequences are provided. The method generates a double strand break (DSB) at a predetermined target site in a genomic DNA sequence, at which target site results in a mutation, insertion and/or deletion of the DNA sequence. Compositions include a DNA construct comprising a nucleotide acid sequence encoding a Cpf1 or Csm1 protein operatively linked to a promoter operative in a cell of interest. The DNA constructs can be used to direct genomic DNA modification at a predetermined genomic locus. Described herein are methods of using these DNA constructs to modify genomic DNA sequences. In addition, compositions and methods for modulating gene expression are provided. Compositions include a DNA construct comprising a promoter operative in a cell of interest operatively linked to a nucleotide sequence encoding a mutant Cpf1 or Csm1 protein that eliminates DSB-producing ability, optionally linked to a domain that modulates transcriptional activity.
