Summary:The present disclosure generally relates to polynucleotides that can be used in calibration methods that determine the identity and/or quantity of polynucleotides in a sample. In particular, the present disclosure relates to polynucleotides comprising a calibration sequence for use as a reference ladder or a reference adaptor. The polynucleotides can be used to calibrate a wide variety of sequencing methods, including high throughput sequencing methods (e.g., those referred to as next generation sequencing or NGS methods).
