The invention provides a purification method of rubber elongation factor protein. The method comprises the following steps: obtaining rubber particles from Brazil hevea brasiliensis latex, and dispersing the rubber particles in a buffer solution; extracting rubber particle protein by using an extracting solution containing a nonionic detergent; adding ammonium sulfate and an alcohol reagent into the extracting solution containing the rubber particle protein to precipitate the rubber particle protein (taking rubber elongation factor protein as a main component); and separating the extracted rubber particle protein through gel filtration chromatography to obtain the purified rubber elongation factor protein.
