The invention provides environment-friendly medical packaging paper as well as a preparation method and application thereof. The preparation method of the packaging paper comprises the following steps: mixing softwood pulp and hardwood pulp, then performing repulping, and then performing pulping treatment to obtain pulp; adding cationic etherified starch, a maleoyl chitosan oxidation derivative and an AKD (alkyl ketene dimer) neutral sizing agent into the pulped pulp, mixing, passing through a paper feeding system, forming by a net part, squeezing and dehydrating, drying, sizing on the surface, drying, calendaring, and manufacturing with paper, so as to obtain the environment-friendly medical packaging paper.
