A method for determining the sensitivity or resistance of at least one identified bacterium to at least one antibiotic, said method comprising the steps of: a) contacting a sample comprising said bacterium with said at least one antibiotic, when said bacterium is "sensitive" to at least one antibiotic, said sample comprising said bacterium; the at least one antibiotic induces rupture of bacterial walls and/or cytoplasmic membranes and causes release of intracellular compounds of the bacteria, b) incubating the sample with the at least one antibiotic, c) purifying the sample by removal of intact bacteria and cell debris, d) analyzing the purified sample by mass spectrometry, and e) determining whether the sample is in the form of the at least one antibiotic.
