Summary:Compositions and methods for modifying genomic DNA sequences are provided. The method produces a double strand break (DSB) at a predetermined target site in the targeted DNA sequence resulting in a mutation, insertion and/or deletion of the DNA sequence at one or more targeted sites. Compositions include a DNA construct comprising a nucleotide sequence encoding a Cpf1 protein operably linked to a promoter operable in a cell of interest. The DNA constructs can be used to direct genomic DNA modification at predetermined locations. Described herein are methods of using these DNA constructs to modify genomic DNA sequences. In addition, compositions and methods for modulating gene expression are provided. Compositions include a DNA construct comprising a promoter operable in a cell of interest operably linked to a nucleotide sequence encoding a mutant Cpf1 protein that eliminates DSB-producing ability, optionally linked to a domain that modulates transcriptional activity.
