The invention relates to hydrophobic polysaccharide microspheres, a preparation method thereof and cosmetics. The method comprises the following steps: providing polysaccharide microspheres prepared from a natural polysaccharide material; and a hydrophobic modification step: contacting a lauroyl lysine solution with the polysaccharide microspheres so that lauroyl lysine is bonded to at least part of the surfaces of the polysaccharide microspheres to obtain the hydrophobic polysaccharide microspheres. After the polysaccharide microspheres are subjected to hydrophobic modification by adopting lauroyl lysine as a hydrophobic modification reagent, the polysaccharide microspheres have hydrophobicity, and the problem of hydrophilicity of common cellulose can be solved, so that the polysaccharide microspheres can be applied to an oil-soluble system.
