The present application relates to compositions and methods for nucleic acid amplification. The present application relates to oligonucleotides for use in various biological assays, including primer extension, nucleic acid amplification, single nucleotide polymorphism detection, and RT-PCR. The oligonucleotide comprises a 5'region complementary to the target sequence and a blocking group at or near the 3 'end of the oligonucleotide.
