The invention relates to a preparation method of hydrogel, in particular to chloroquine drug induced tough hydrogel and a preparation method and application thereof, the preparation method comprises the following steps: (1) dissolving biomacromolecules in water to form a biomacromolecule solution; (2) adding chloroquine drugs into the biomacromolecule solution, uniformly mixing, and standing; (3) after an amyloid polymer is generated in the biomacromolecule solution, heating and stirring to form prototype hydrogel; and (4) transferring the prototype hydrogel into a vacuum humidity control device, and removing bound water to obtain the hydrogel.
