The invention relates to a preparation method of hydrogel, in particular to hydrogel embedded with chloroquine medicine as well as a preparation method and application thereof, the preparation method comprises the following steps: (1) adding chloroquine medicine into a liquid dairy product, uniformly mixing, and performing phase separation on the liquid dairy product to obtain a mixed solution; (2) shearing and homogenizing the mixed solution to obtain a homogenized solution; (3) centrifuging the homogenized liquid, removing supernatant and unreacted chloroquine drugs in the supernatant after centrifugation is completed, and taking precipitate for later use; and (4) reducing the precipitation temperature to form gel, so as to obtain the hydrogel embedded with the chloroquine medicine.
