The preparation method comprises the following step: carrying out cyclization reaction on 3-oxopropionitrile and guanidine under an oxidation condition to generate the 2, 4-diaminopyrimidine oxide. Wherein the 3-oxopropionitrile is obtained by oxidizing a medicinal chemical 3-hydroxypropionitrile which is low in price and easy to obtain and is used as a raw material.
