Summary:The invention discloses a method for inducing an endophyte-free tender shoot by using plant branches, and belongs to the field of plant tissue culture. The method comprises the following steps that (1), branches of more than biennial plants are selected, thoroughly washed with tap water, and aired; (2), the bottom ends of the aired plant branches are soaked in a growth solution, standing is carried out for 1-3 hours, and the growth solution is prepared by dissolving 1-naphthylacetic acid and ethanol according to the ratio of (0.5-2.5 g): (100-300 mL) and then diluting with distilled water; and (3), a culture solution is prepared, ampicillin and a gentamicin solution are added, the branches soaked in the step (2) are transferred into the culture solution, and illumination/darkness alternative closed culture is carried out to obtain the endophyte-free tender shoot.
