The method comprises the following steps: (1) carrying out subcritical extraction on a tobacco leaf sample to obtain a tobacco crude extract, carrying out alcohol precipitation and impurity removal on the tobacco crude extract to obtain an extracting solution, and then concentrating the extracting solution to obtain a concentrate; and (2) carrying out molecular distillation separation on the concentrate at the temperature of 40-80 DEG C under the pressure of 0.001-0.01 mbar, and removing light components such that residual heavy components are the tobacco extract.
