The invention relates to a preparation method of a graphene disinfectant. The preparation method comprises the following sequential steps: 1, pouring graphene powder into water, adding a dispersing agent, and conducting uniform stirring to obtain a graphene pre-dispersion liquid; 2, placing the obtained graphene pre-dispersion liquid in a microwave reactor for treatment for 4-8 min so as to obtain a graphene suspension, and carrying out ultrasonic treatment for 1-2 h to obtain a homogeneous and stable graphene dispersion liquid; 3, pouring the stable graphene dispersion liquid into a reaction kettle, adding a nonionic surfactant into the reaction kettle, carrying out stirring and mixing, then adding quaternary ammonium salt, and carrying out mixing and stirring; 4, detecting the concentration of a graphene pre-disinfectant at the moment, and performing fine adjustment; and 5, stirring the graphene pre-disinfectant again to enable the graphene pre-disinfectant to undergo sufficient reaction so as to obtain the
