The invention discloses a method for preparing fish actin by utilizing an aqueous two-phase method and application thereof. The method for preparing the fish actin by utilizing the aqueous two-phase method comprises the following steps: mixing short-chain alcohol, salt and water to obtain an aqueous two-phase system; adding fish fibrillin or a fish fibrillin solution into the aqueous two-phase system, and carrying out uniform mixing to obtain an aqueous two-phase separation and extraction system; and then, waiting until the aqueous two-phase separation and extraction system is layered again, and obtaining the upper phase to obtain a fish actin solution. The method further comprises a step of quantifying the fish actin by using a capillary tube. The method disclosed by the invention is highin extraction rate and high in product purity, so that the method can be used for fish actin production or fish actin standard substance preparation. 本发明公开了一种利用双水相法制备鱼类肌动蛋白的方法及其应用。本发明通过将短链醇、盐和水混合得到双水相体系；然后在双水相体系中加入鱼肉肌原纤维蛋白或是
