Summary: | The cultivation includes the following processes successively: 1. Before blooming, tender ephedra intermedia branches as inoculation material are cut down, washed and disinfected with 0.2% mercury chloride solution for 8 minutes; 2. Tender branch sections are dark-cultured in inducible culture medium inside growth cabinet; 3. Embryonal callus produced during inducible culture is cut down for two-step subculture; 4. EC is placed in LC subculture medium for the first step of subculture; and 5. Yellowish EC produced is placed in LC medium for the second step of subculture. The said method can retard the degeneration of culture and ephedrine content and obtain more callus and high ephedrine content. |
