Summary: | The invention discloses a high-throughput detection method for the content of ergothioneine in a to-be-detected sample and application of the high-throughput detection method, and belongs to the technical field of microbial breeding. The method comprises the following steps: adjusting the pH value of a to-be-detected sample to 1.9-2.0, firstly adding a ferric thiocyanate solution into an elisa plate, then adding the to-be-detected sample, and conducting blowing, beating and mixing; and recording the absorbance by using a microplate reader, wherein the wavelength used by the microplate reader is 450nm-470nm. The lower the absorbance is, the higher the ergothioneine content is. The method has the characteristics of high efficiency, rapidness, convenience and low cost, and is suitable for research activities which do not require accurate determination of the ergothioneine content of each sample; and the defects of time consumption and high cost of an LCMS detection method are overcome.
