The method comprises the following steps: (1) resuspending PBMC cells in a culture container with a 1640 culture solution, and then placing the culture container in an incubator with constant temperature and a gas carbon source for culture; (2) shaking the culture container after culturing for a predetermined time, discarding suspended cells, then adding a DC serum-free medium, a GM-CSF reagent, an Il-4reagent and a cholesterol reagent, and continuing culturing in the incubator; and (3) adding LPS and IL-1 alpha when culturing for the third day, and continuing culturing until mature DC cells are obtained.
