Summary: | The invention provides an SNP analysis technique based on NGS, for detecting brain gliomas 1p and 19q chromosomes. The technique comprises the following steps of step (1), extracting DNA of a sample to be detected, and performing quantitative detection on the concentration by Qubit 3.0, wherein the sample with the qualified concentration is used as a high-flux sequencing formwork; step (2) after genomic DNA is interrupted, performing end repair; step (3), purifying connector connection and connector connection products; step (4), after a pre-library is amplified, purifying the amplified pre-library; step (5), performing hybridization on the pre-library; and step (6), performing catching, eluting, preparing an end library and purifying the end library. According to the SNP analysis technique method based on NGS, 1p and/or 19q deletion can also be reliably detected in background tumor tissue of 70% of normal cells, and the method is more sensitive than LOH analysis based on microsatellite. Less DNA is needed, a |
