The preparation method comprises the steps that 1, cornstraw is crushed and stacked, complex microbial inoculants are scattered and fermented, urea is added, water is added finally, and stacking is conducted; 2, fermentation is conducted for three days,fermentation continues to be conducted while turning is conducted, fermentation is conducted till the fermentation temperature is lowered to the room temperature, the corn straw obtained after thorough decomposition is screened with a sieve, and the bioorganic substrate special for the virus-free potato breeder's seeds is obtained. The method has the advantages that comprehensive utilization of the corn straw is achieved,
