The invention provides a determination method and device for an organic acid yield of organic matter in an underground hydrocarbon source rock stratum. The method comprises the steps that feature dataof organic geochemistry of the hydrocarbon source rock stratum is acquired; a relation curve of the acid yield and organic matter maturity in the hydrocarbon source rock stratum is established, and the acid yield is acquired; organic matter maturity in the hydrocarbon source rock stratum at different depths is subjected to simulating calculation; original organic carbon content in the hydrocarbonsource rock stratum is calculated; the produced acid strength of a single type of organic matter in the hydrocarbon source rock stratum is calculated; and a total organic acid yield of all types of organic matter in the hydrocarbon source rock stratum is calculated.
